Have you tried it without [] like
update [$_temptable] set col3 = #count where col1SQL = $building
or
update [$_temptable] set col3 = #count where col1SQL = $_building
if it's a global variable...
also you could try
move 'where col1SQL = ' || $building to $where
update [$_temptable]
set col3 = #count
[$where]
